I'm a new PS3 owner and would like to know if the system can play a movie file that's roughly 4.6GB in size. Now, I do understand that the PS3 can only read FAT or FAT32 formatted USB HD's right?

If I can transfer this to a NTFS USB flash stick (on my PC), then re-format this to FAT or FAT32 (while retaining the data which I think is possible), and then stick it in my PS3, can I watch this 720p quality movie??

This would really help if someone can provide a thorough answer to this, thanks. Will +rep.

No.
If you change the file structure, the same limits apply. And 4GB is the maximum single file size for FAT32.
You won't be able to use your suggested trick.... it probably won't even allow for a file structure change.
